Mediators and Moderators of Treatment Effects in RCTs (Geelong)

Randomised clinical trials (RCTs) not only are the gold standard for evaluating the efficacy and effectiveness of treatments but also can be used in examining moderators and mediators of the response. In the RCT setting, moderators identify on whom and under what circumstances treatments have different effects, and mediators identify why and how treatments have effects. Because an effective intervention is not equally effective for all members of a given population, establishing moderators of intervention response can prompt researchers to optimally target the intervention, and to seek better interventions for non-responders. Establishing mediators of intervention response can prompt researchers either to strengthen, add, or remove certain intervention components to make the intervention either more efficacious or more cost-effective.

Given the increasingly important role of moderators and mediators in clinical research, clear definitions of the two terms are essential to avoid inconsistent, ambiguous, and possibly misleading results. We will describe an analytic framework to identify and distinguish between moderators and mediators in RCTs, we will then review and integrate the methodological literature concerning dealing with covariates. Statistical methods for examining mediators and moderators in the RCT setting using linear mixed models will be also discussed.

Moderators, mediators and predictors of outcomes using RCTs in psychiatric research will be discussed as case studies.

Who should attend?

PhD students and early career researchers within the Faculty of Health.

Participants should be familiar with RCT design. The workshop assumes knowledge of statistics and regression models. Understanding basic concept of linear mixed models for analysing RCTs would be beneficial.
